Where the Melatonin Itself Comes From

The melatonin in the vast majority of supplements is synthetic, built from chemical precursors in a factory rather than harvested from a biological source. The end product is chemically identical to the melatonin your brain produces at night. Synthetic production allows manufacturers to create large, consistent batches at low cost, which is why it dominates the market.

A smaller category of supplements uses plant-derived melatonin, sometimes labeled “phytomelatonin.” These products typically come from dried extracts of alfalfa, chlorella algae, and rice. The melatonin molecule itself is structurally the same whether it’s synthetic or plant-derived. The difference is that plant extracts carry along other compounds from the source material: chlorophyll, beta-carotene, isoflavones, and various plant metabolites. Research published in the journal Molecules found that one phytomelatonin extract was five to ten times more effective as an antioxidant in lab tests than synthetic melatonin at comparable concentrations, likely because of these additional plant compounds rather than any difference in the melatonin itself.

A third production method is emerging but not yet widespread in consumer products. Scientists have engineered bacteria (E. coli) and yeast (the same species used in beer and wine) to produce melatonin through fermentation. These microorganisms are modified to carry genes that convert the amino acid tryptophan into serotonin and then into melatonin, mimicking the same biological pathway your body uses. This bio-manufacturing approach is gaining interest from pharmaceutical companies, but most pills on store shelves still use the traditional chemical synthesis route.

From Powder to Pill

Whether the melatonin is synthetic or plant-derived, it arrives at the pill manufacturer as a fine powder. That powder alone can’t become a functional tablet. A typical 3 mg or 5 mg melatonin dose is a tiny amount of material, far too small to pick up with your fingers, so the bulk of every pill is actually made up of inactive ingredients that give it size, shape, and structural integrity.

A standard melatonin tablet contains microcrystalline cellulose as its primary filler, a plant-based fiber that forms the body of the pill. Magnesium stearate acts as a lubricant so the powder doesn’t stick to the machinery during pressing. Silica (silicon dioxide) keeps the powder flowing smoothly through equipment. Croscarmellose sodium is a disintegrant: it swells when it contacts moisture in your stomach, helping the tablet break apart so the melatonin can absorb. Many tablets also get a thin coating made from hypromellose, polyethylene glycol, and hydroxypropyl cellulose, which makes them easier to swallow and protects the melatonin from light and moisture degradation.

The manufacturing steps follow a standard supplement production process. The melatonin powder is weighed and blended with the inactive ingredients in precise ratios using industrial mixers. The blended powder is then fed into a tablet press, a machine that compresses it under high pressure into uniform pill shapes. Coated tablets go through an additional step in a rotating drum where the coating solution is sprayed on and dried.

Gummies, Liquids, and Quick-Dissolve Formats

Not all melatonin comes as a pressed tablet. Gummies are made by dissolving melatonin into a heated mixture of gelatin (or pectin for vegan versions), sweeteners, flavoring, and coloring agents. This liquid is poured into molds, cooled, and often coated with a light layer of sugar or citric acid. The challenge with gummies is that melatonin can degrade when exposed to heat during manufacturing, which is one reason independent lab tests frequently find that gummy melatonin content doesn’t match what’s on the label.

Liquid melatonin is dissolved in a solvent base, often glycerin or water with preservatives and flavoring. Quick-dissolve tablets use a different compression technique that creates a more porous structure, allowing the pill to disintegrate on your tongue within seconds rather than needing stomach acid to break it apart. Extended-release formulations add a polymer matrix or coating that slowly erodes, releasing melatonin over several hours to mimic your body’s natural production pattern.

Quality Control and Purity Concerns

Because melatonin is sold as a dietary supplement in the United States rather than a regulated drug, manufacturers are not required to prove their products contain what the label claims before going to market. This creates real variability. Independent testing has repeatedly found that actual melatonin content in supplements can range from a fraction of the labeled dose to several times higher.

During synthesis, unwanted chemical byproducts can form. Pharmaceutical-grade standards, like those set by the German Drug Codex, require that any individual impurity stay below 0.1% of total content and all impurities combined stay below 0.5%. Melatonin also degrades when exposed to acidic or alkaline conditions, hydrogen peroxide, or UV light, producing breakdown products that are difficult to fully identify with standard testing methods. This is why storage matters: heat, light, and humidity all accelerate degradation.

Manufacturers who follow Good Manufacturing Practices (GMP) test raw melatonin powder for identity and purity before production, then test finished pills for content uniformity, meaning each pill in a batch should contain the same amount. Third-party certifications from organizations like USP or NSF International provide an additional check, verifying that what’s on the label matches what’s in the bottle.
