The drug reaches its highest concentration in your bloodstream roughly 2 hours after you take it. That’s when you’re getting the full pharmacological effect. So if you know you’ll be sexually active in the evening, taking your dose in the late afternoon gives you the best chance of peak performance when you want it.

What sets tadalafil apart from other erectile dysfunction medications is how long it lasts. The drug has a half-life of about 17.5 hours, which translates to a functional window of up to 36 hours. That means a single dose taken on Friday afternoon could still be working Sunday morning.
