A smoked or vaporized DMT experience typically lasts about 15 to 20 minutes from first effects to near-complete resolution, making it one of the shortest psychedelic trips in existence. That figure changes dramatically depending on how the substance enters the body: an intravenous injection produces a similar window, while oral DMT combined with a monoamine oxidase inhibitor (the basis of ayahuasca) stretches the experience to four to six hours. The route of administration is the single biggest variable, and understanding why reveals quite a bit about how the molecule works once it’s in your system.
Smoked and Vaporized DMT
When DMT is inhaled as vapor, it crosses from the lungs into the bloodstream almost instantly. Psychedelic effects peak within roughly two to five minutes and resolve within about 20 minutes.1UTHealth Houston. Vaporized DMT Shows Promise for Rapid and Sustained Relief in Treatment-Resistant Depression Users often describe the onset as abrupt, sometimes overwhelming, with vivid visual imagery and a sense of being transported to an entirely different environment. The brevity of the experience is sometimes called a “breakthrough” trip, and many people report that despite clocking in under half an hour, the subjective sense of time can feel much longer. The return to baseline is equally rapid: within 30 minutes of inhalation, most people feel close to normal.
Intravenous DMT
In clinical research settings, DMT has been administered as a direct injection into a vein. The timeline closely mirrors smoked DMT. Psychological effects begin almost immediately after the injection, peak at about 90 to 120 seconds, and are nearly completely resolved by 30 minutes.2JAMA Psychiatry. Dose-Response Study of N,N-Dimethyltryptamine in Humans: II. Subjective Effects and Preliminary Results of a New Rating Scale Peak blood levels of DMT also appear at about two minutes post-injection and drop off steeply from there.3Translational Psychiatry. Acute effects of intravenous DMT in a randomized placebo-controlled study in healthy participants So whether you smoke it or inject it, the body processes DMT on roughly the same compressed schedule.
The intravenous route does offer researchers one advantage: precise control over dosing. In the studies that established the 30-minute window, blood levels of DMT tracked tightly with subjective intensity, falling to negligible concentrations at the same point people reported feeling sober again.4Brazilian Journal of Psychiatry. Why DMT Disappears So Fast
DMT’s brevity comes down to how aggressively the body breaks it down. The enzyme primarily responsible is monoamine oxidase A (MAO-A), which sits in high concentrations in the liver and gut. Once DMT hits the bloodstream, MAO-A converts the vast majority of it into an inactive byproduct called indole-3-acetic acid. In one study comparing smoked and oral DMT metabolism, that single byproduct accounted for about 97% of all recovered DMT-related compounds in urine, with only about 3% turning into a secondary metabolite called DMT-N-oxide.5PubMed. Metabolism and urinary disposition of N,N-dimethyltryptamine after oral and smoked administration: a comparative study A small fraction of DMT is also processed by certain liver enzymes from the cytochrome P450 family, but MAO-A does the lion’s share of the work.6PubMed Central. Clinical Pharmacokinetics of N,N-Dimethyltryptamine (DMT): A Systematic Review and Post-hoc Analysis
This enzymatic efficiency explains a key pharmacokinetic feature. In a placebo-controlled trial measuring blood levels during and after an intravenous infusion, researchers observed two distinct half-lives once the infusion stopped: an early one averaging five to six minutes and a later one averaging 14 to 16 minutes. About three-quarters of the DMT was cleared during that first, faster phase.7Translational Psychiatry. Acute effects of intravenous DMT in a randomized placebo-controlled study in healthy participants In practical terms, the molecule is mostly gone before you’d have time to settle into a long conversation about the experience.
Oral DMT and Ayahuasca
If you swallow pure DMT by itself, almost nothing happens. MAO-A in the gut lining and liver destroys the molecule before it ever reaches the brain in meaningful quantities. This is why ayahuasca, the traditional Amazonian brew, combines DMT-containing plants with plants rich in harmala alkaloids. These alkaloids are monoamine oxidase inhibitors: they temporarily block the enzyme that would otherwise neutralize DMT on first pass through the gut. The result is a dramatically longer experience, typically lasting four to six hours.8Brazilian Journal of Psychiatry. The pharmacological interaction of compounds in ayahuasca: a systematic review
The character of the ayahuasca experience also differs from a smoked DMT trip. The onset is slower, usually taking 30 to 60 minutes as the DMT absorbs through the stomach and intestines. The peak tends to be less explosively intense but far more sustained, with gradual waves of visual and emotional content. For someone used to the compressed intensity of inhaled DMT, the shift to a multi-hour oral experience is striking. The trade-off is time: you are committing a large chunk of your day, and nausea is common during the come-up, a side effect largely absent from other routes.
Pharmahuasca and Synthetic Oral Formulations
Researchers have also studied “pharmahuasca,” which replaces the plant-based brew with measured pharmaceutical doses of DMT and a purified MAO inhibitor like harmine. In a dose-escalation study of 16 healthy participants who received DMT combined with harmine via a transmucosal delivery system, all combinations produced subjective effects lasting four to five hours, with peak blood levels scaling up predictably with dose.9PubMed. Examining the pharmacokinetic and pharmacodynamic interaction of N,N-dimethyltryptamine and harmine in healthy volunteers: Α factorial dose-escalation study The duration is comparable to traditional ayahuasca, which makes sense: the underlying mechanism (MAO inhibition allowing oral DMT absorption) is the same whether the ingredients come from a rainforest vine or a chemistry lab.
Extending the Trip With Continuous Infusion
One limitation of inhaled or bolus-injected DMT for clinical use is that the experience may be too short for therapeutic work. By the time a person begins to orient themselves in the psychedelic state, it’s already fading. This has driven research into continuous intravenous infusion, sometimes called “DMT drip” protocols, which sustain steady blood levels for a chosen duration.
In one study, researchers gave an initial bolus injection followed by a constant-rate infusion. Subjective effects peaked within the first two minutes of the bolus, dipped slightly, and then stabilized at a plateau that held throughout a 30-minute infusion. Effects remained elevated for about 12 minutes after the infusion stopped.10PubMed Central. Psychological and physiological effects of extended DMT A more ambitious trial pushed the infusion window to six hours, demonstrating that it was safe and tolerable to maintain a psychedelic state for that length of time in healthy volunteers.11PubMed Central. Safety, Pharmacokinetics, and Pharmacodynamics of a 6-h N,N-Dimethyltryptamine (DMT) Infusion in Healthy Volunteers: A Randomized, Double-Blind, Placebo-Controlled Trial Once the infusion stops, blood levels plummet within seconds: the same study measuring half-lives found that DMT concentrations dropped very rapidly, within 15 to 20 seconds, once the drip ended.12Translational Psychiatry. Acute effects of intravenous DMT in a randomized placebo-controlled study in healthy participants This gives clinicians something unusual for a psychedelic: an off switch.
How the Body Recovers Physically
DMT doesn’t just produce visual and psychological effects. It also temporarily raises blood pressure and heart rate. These physiological changes follow the same compressed timeline as the subjective experience. After an intravenous bolus, blood pressure and heart rate peak within about two minutes and then return to normal levels. When DMT is delivered as a slower infusion, the cardiovascular effects are milder, only slightly elevated compared to placebo, and they normalize within about 15 minutes of stopping the drip.13Translational Psychiatry. Acute effects of intravenous DMT in a randomized placebo-controlled study in healthy participants – Section: Autonomic and adverse effects A separate dose-ranging study in both healthy volunteers and people with major depression found that increases in blood pressure, heart rate, and anxiety resolved within 20 to 30 minutes of injection.14PubMed Central. Exploratory study of the dose-related safety, tolerability, and efficacy of dimethyltryptamine (DMT) in healthy volunteers and major depressive disorder
For most healthy people, the physical after-effects are minimal once the acute window closes. There’s no hangover in the traditional sense. Some people describe feeling emotionally raw or reflective for hours afterward, but from a cardiovascular and autonomic standpoint, the body seems to reset quickly.
Does Tolerance Build Up Between Doses?
With most classic psychedelics, repeated doses over a short period lead to rapidly diminishing effects, a phenomenon called acute tolerance or tachyphylaxis. DMT is unusual in this regard. In a study where participants received four closely spaced intravenous doses of DMT, tolerance to the psychedelic effects did not develop: people reported similar intensity across all four doses, whether measured by clinical interview or standardized rating scales. Interestingly, some of the body’s hormonal responses (like cortisol and stress hormones) did diminish with repeated dosing, and heart rate responses also decreased, even as the subjective experience stayed strong.15Biological Psychiatry. Differential tolerance to biological and subjective effects of four closely spaced doses of N,N-dimethyltryptamine in humans
This disconnect between subjective and physiological tolerance is genuinely unusual among psychedelics and is still not fully understood. It may relate to the way DMT interacts with its receptor targets differently from compounds like psilocybin or LSD, or it may reflect DMT’s rapid clearance preventing the kind of sustained receptor occupancy that triggers tolerance. Whatever the explanation, it means that back-to-back DMT sessions don’t fade the way back-to-back psilocybin sessions would.
Psychological Aftereffects That Outlast the Trip
The acute psychedelic experience may last 20 minutes or a few hours, but the psychological impact can persist far longer. In a placebo-controlled study of healthy volunteers who received intravenous DMT, significant reductions in depression scores were still present one week later, and in a broader prospective sample, those reductions held at two weeks. The degree of improvement correlated with the intensity of a particular aspect of the experience often described as oceanic boundlessness, a feeling of unity or transcendence.16Scientific Reports. Effects of DMT on mental health outcomes in healthy volunteers
Research on 5-MeO-DMT (a structurally related but distinct compound found in certain toad secretions) paints a similar picture. In a naturalistic study following people who inhaled 5-MeO-DMT, ratings of life satisfaction and convergent thinking increased immediately after the session and remained elevated four weeks later. Depression, anxiety, and stress scores decreased and reached statistical significance at the four-week mark.17PubMed Central. A single inhalation of vapor from dried toad secretion containing 5-methoxy-N,N-dimethyltryptamine (5-MeO-DMT) in a naturalistic setting is related to sustained enhancement of satisfaction with life, mindfulness-related capacities, and a decrement of psychopathological symptoms These are self-reported outcomes from a naturalistic setting rather than a controlled trial, so the results deserve some caution. But the general pattern, a brief psychedelic window followed by weeks of measurable psychological change, appears consistently across multiple studies and compounds in this family.
5-MeO-DMT Is Not the Same Drug
People sometimes use “DMT” loosely to mean either N,N-DMT (the compound found in ayahuasca) or 5-MeO-DMT (the compound from Sonoran Desert toad secretions and certain plants). These are pharmacologically distinct molecules with different receptor profiles and different subjective qualities. 5-MeO-DMT tends to produce less visual content and more intense feelings of ego dissolution and emotional amplification. But the duration profile is broadly similar: when smoked or insufflated, 5-MeO-DMT is short-acting, with a rapid onset and effects that generally resolve within 30 to 45 minutes.18PubMed Central. The clinical pharmacology and potential therapeutic applications of 5-methoxy-N,N-dimethyltryptamine (5-MeO-DMT) If someone tells you their “DMT trip” lasted 40 minutes and felt more like dissolution than visuals, they may have taken 5-MeO-DMT rather than N,N-DMT.
Intranasal and Other Emerging Routes
Researchers are exploring routes beyond smoking, drinking, and injection. Intranasal delivery (essentially a nasal spray) is attracting attention because it could offer a non-invasive, clinic-friendly option. In animal studies, intranasal DMT showed faster pharmacokinetics than subcutaneous injection, with a half-life in the range of 12 to 14 minutes compared to a much longer and more variable 45 to 123 minutes for the subcutaneous route.19PubMed Central. Pharmacodynamic effects and plasma pharmacokinetics of N,N-dimethyltryptamine after intranasal versus subcutaneous administration in male rats These are rat data, so the exact numbers won’t translate directly to humans, but the direction is interesting: intranasal delivery appears to behave more like inhaled DMT (fast in, fast out) than like a slower absorption route.
A proof-of-concept trial using intranasal 5-MeO-DMT in people with treatment-resistant depression who were already on SSRIs found that participants were assessed as ready for discharge from the clinic roughly 96 to 102 minutes after dosing, depending on the dose cohort.20PubMed Central. Intranasal 5-MeO-DMT Concomitant with SSRI for Treatment-Resistant Depression: A Proof-of-Concept Trial That two-hour observation window before discharge gives a practical sense of the clinical recovery timeline, even though the peak psychedelic effects would have subsided earlier.
Why Duration Varies So Much Between People
Even within a single route of administration, individual variation exists. Factors that influence how long DMT effects last include dose (higher doses tend to produce somewhat longer and more intense experiences), individual differences in MAO-A enzyme activity, body composition, and whether any other substances are present. Someone taking an antidepressant that partially inhibits MAO, for instance, could experience a longer-than-expected trip because the enzyme that clears DMT isn’t working at full capacity. Anything that slows the liver’s ability to process the molecule, whether a drug interaction or a genetic variation in enzyme expression, has the potential to extend the duration.
Set and setting also matter for the subjective sense of how long the trip feels, even if they don’t change the pharmacokinetics. People in unfamiliar or anxiety-provoking environments often report that the experience feels longer than it is by the clock. Someone who has smoked DMT before and knows what to expect may perceive the same 15-minute trip as shorter than a first-time user would. The clock time stays the same, but the psychological experience of duration is elastic.
A Quick Reference by Route
Because the duration question depends so heavily on how DMT enters the body, here’s a straightforward comparison:
- Smoked or vaporized: Effects peak in 2 to 5 minutes; most people feel baseline within 15 to 20 minutes, with full resolution by 30 minutes.
- Intravenous bolus: Peak at about 90 seconds; nearly resolved by 30 minutes.
- Intravenous infusion: Effects sustained for the length of the drip, which in research has ranged from 30 minutes to 6 hours, with rapid return to baseline once the infusion stops.
- Oral with MAO inhibitor (ayahuasca or pharmahuasca): Onset in 30 to 60 minutes; total duration of 4 to 6 hours.
- Oral without MAO inhibitor: Essentially inactive. MAO-A in the gut destroys DMT before absorption.
The physiological effects, primarily elevated blood pressure and heart rate, track closely with the subjective timeline and generally normalize within 15 to 30 minutes of the experience ending, regardless of route.
