How Long Does Wellbutrin Stay in Your System?

Wellbutrin (bupropion) takes roughly 4 to 5 days for the parent drug to clear your body after your last dose, but its active metabolites linger longer. Because some of these breakdown products have half-lives of 33 to 36 hours, full elimination of all active compounds typically takes about 8 to 10 days for most people.

How the Half-Life Determines Clearance

Bupropion itself has a mean elimination half-life of about 21 hours after regular dosing. That means roughly half the drug is gone every 21 hours. After four to five half-lives, a substance is considered effectively eliminated, which puts bupropion clearance at about 4 to 5 days.

But bupropion doesn’t just disappear. Your liver breaks it into three active metabolites that are still pharmacologically active, meaning they continue affecting your brain chemistry. Hydroxybupropion has a half-life of around 20 hours, similar to the parent drug. The other two metabolites, threohydrobupropion and erythrohydrobupropion, hang around considerably longer with half-lives of 33 to 36 hours. These slower-clearing metabolites are the reason it takes over a week for all traces of Wellbutrin’s activity to leave your system.

Formulation Doesn’t Change Total Clearance Time

Wellbutrin comes in three formulations: immediate-release (IR), sustained-release (SR), and extended-release (XL). The XL version absorbs more slowly, reaching peak blood levels around 5 hours after a dose compared to about 1.5 hours for IR. However, all three formulations are bioequivalent, meaning they deliver the same total amount of drug to your body. The formulation affects how quickly the drug gets in, not how long it takes to get out. Your total clearance timeline is essentially the same regardless of which version you take.

Liver and Kidney Problems Slow Elimination

Your liver does the heavy lifting in breaking down bupropion, so liver disease can significantly extend clearance times. In people with severe liver cirrhosis, the half-life of bupropion itself stretches from about 19 hours to 29 hours. More notably, the half-life of hydroxybupropion increases fivefold, and the other two metabolites double their half-lives. That could push full elimination well past two weeks.

Kidney impairment also matters. In people with end-stage kidney disease, levels of the active metabolites build up to 2 to 3 times higher than normal, because the kidneys can’t clear them efficiently. Even moderate kidney impairment roughly doubles overall drug exposure from a single dose.

The effect of age is less clear-cut. FDA data shows no consistent relationship between age and bupropion blood levels in adults 18 to 83. One study found that single-dose clearance looked similar in older and younger adults, but another suggested elderly patients may accumulate more of the drug and its metabolites over time with repeated dosing.

What Discontinuation Feels Like

Wellbutrin has a lower risk of discontinuation symptoms compared to many other antidepressants, but some people do notice effects as the drug leaves their system. The timeline generally follows a predictable pattern.

During the first 1 to 3 days, most people feel fine as blood levels drop gradually. Some notice mild brain fog or low energy. Days 4 through 7 are when symptoms tend to peak if they appear at all. This can include feeling scattered or unusually tired, headaches, muscle aches, sleep disruption, mood shifts, or cravings. By the second week, most symptoms have faded as the nervous system readjusts. Lingering effects beyond three weeks are rare.

The Two-Week Washout for MAOIs

If you’re switching from Wellbutrin to a monoamine oxidase inhibitor (MAOI), clinical guidelines call for a two-week washout period after your last dose of bupropion. This buffer ensures the drug and its long-lasting metabolites have fully cleared before starting a medication that could interact dangerously with any remaining bupropion activity. The same two-week window applies in reverse when switching from an MAOI to bupropion.

False Positives on Drug Tests

One practical concern many people have is whether Wellbutrin can affect drug test results. It can. Bupropion and two of its metabolites are structurally similar to amphetamine, which causes cross-reactivity on common urine immunoassay screens. In one emergency department study, bupropion was responsible for 41% of amphetamine results that turned out to be false positives on confirmatory testing. This isn’t limited to one testing method either; false positives have been documented across multiple immunoassay platforms.

If you’re taking Wellbutrin and face a urine drug screen, the initial result may flag positive for amphetamines. A confirmatory test using a more precise method (gas chromatography-mass spectrometry) will distinguish bupropion from actual amphetamines. Informing the testing facility that you take bupropion beforehand can save time and stress. Because the metabolites take over a week to clear, this false-positive risk persists for roughly 8 to 10 days after your last dose.
