A 500mg dose of vitamin B1 (thiamine) is not considered toxic, but most of it will pass straight through your body without being absorbed. The recommended daily intake for adults is just 1.1 to 1.2 mg, making 500mg roughly 400 times that amount. No official upper limit exists for thiamine because no adverse effects have been reliably documented at high oral doses, but that doesn’t mean taking 500mg is the same as taking a useful amount.

Your body simply stops pulling it in from the gut and lets the rest pass through.
That said, the Food and Nutrition Board added a caveat. The absence of reported adverse effects doesn’t guarantee that excessive intakes are harmless. It may just mean that so little gets absorbed at extreme doses that problems rarely develop.
What Your Body Actually Absorbs
Thiamine absorption is an active process that saturates quickly. Research on oral dosing found that increasing a dose from 2.5mg to 20mg only added about 0.2mg of extra thiamine in urinary excretion, meaning the gut hit its absorption ceiling early. At 500mg, the vast majority of the thiamine you swallow never enters your bloodstream. It continues through your digestive tract and is eliminated.
Interestingly, splitting doses improves absorption significantly. One study found that taking 10mg as four separate 2.5mg doses spaced two hours apart resulted in three times more thiamine absorption than taking the full 10mg at once. If you’re supplementing for a specific reason, smaller divided doses are far more efficient than one large pill.
Your kidneys also play a role in clearing excess thiamine from the blood. Specialized transporters in the kidney tubules reabsorb thiamine to keep levels stable, but when blood levels are high, the surplus is filtered out into urine. This is why high-dose B vitamins often turn urine bright yellow.
When High Doses Are Used Clinically
Doctors do prescribe high-dose thiamine for specific conditions, particularly alcohol use disorder, where chronic drinking impairs thiamine absorption and kidney reabsorption. Severe deficiency can cause Wernicke’s encephalopathy, a potentially life-threatening brain condition, and aggressive supplementation is part of standard treatment. In these cases, intravenous thiamine is typically used because oral absorption is too limited and unreliable at the doses needed.
People with diabetes may also have lower thiamine levels. Research has found that high blood sugar can reduce the expression of thiamine transporters in the kidneys, causing the body to lose more thiamine through urine than it normally would. Some clinicians recommend supplementation for these patients, though typically at more moderate doses than 500mg.
The Form of B1 Matters
Standard thiamine supplements use thiamine hydrochloride, the water-soluble form with the absorption ceiling described above. A different form called benfotiamine is fat-soluble and bypasses that bottleneck entirely. It crosses intestinal cell membranes passively rather than relying on the transporters that saturate at low doses.
The difference in absorption is dramatic. In a pharmacokinetic study published in The Journal of Clinical Pharmacology, benfotiamine delivered peak blood levels of thiamine that were over 18 times higher than the same dose of thiamine hydrochloride. Total thiamine exposure in the blood was roughly 50 times greater. This matters because if you’re taking 500mg of standard thiamine hydrochloride hoping to get high blood levels, you’re wasting most of it. A much smaller dose of benfotiamine would deliver more usable thiamine to your cells.
No accumulation of breakdown products was observed with repeated benfotiamine dosing, which is a reassuring safety signal. But because benfotiamine actually gets absorbed in meaningful quantities at higher doses, the question of “too much” becomes more relevant with this form than with standard thiamine.
Practical Risks of 500mg
For most people, taking 500mg of thiamine hydrochloride daily is unlikely to cause harm. The body’s absorption limit and kidney clearance act as a double safety net. You’re essentially paying for 500mg and absorbing a tiny fraction of it.
That said, “no documented toxicity” is not the same as “proven safe at any dose.” The research base for very high oral thiamine doses is limited, not because studies have confirmed safety, but because few large trials have specifically tested for subtle long-term effects. Rare allergic reactions to thiamine supplements have been reported, though these appear to be related to the compound itself rather than the dose.
If you’re taking 500mg because you think more is better, the absorption data suggests otherwise. Your gut can only pull in a few milligrams at a time regardless of how large the dose is. If you’re taking it under medical guidance for a diagnosed deficiency or a specific condition, the risk profile is generally considered acceptable given the clinical need. For general wellness, a dose in the 25 to 100mg range delivers everything your body can realistically use from a single dose, without the waste.
