The three vitamins that matter most for overall health are vitamin D, vitamin B12, and vitamin C. These three stand out because they each govern a body system you can’t function without: bone integrity and immune defense, nerve signaling and blood cell production, and tissue repair. They’re also among the most common deficiencies worldwide, making them the vitamins most likely to cause real problems if you’re not getting enough.
Vitamin D: Your Immune and Bone Regulator
Vitamin D does far more than keep your bones strong. Your body converts it through two steps, first in the liver and then in the kidneys and immune cells, into an active hormone that switches on genes throughout the immune system. Receptors for this hormone sit on nearly every type of immune cell, including the T cells and macrophages that form your front line against infection.
On the immune side, vitamin D triggers production of natural antimicrobial compounds that fight bacteria, viruses, and fungi. It also boosts the ability of macrophages to engulf and destroy pathogens while simultaneously dialing down inflammatory signals like IL-6 and TNF-alpha, two molecules responsible for the out-of-control inflammation seen in autoimmune diseases. At the same time, it promotes regulatory T cells, which act as peacekeepers that prevent your immune system from attacking your own tissues.
For bones, vitamin D controls how much calcium and phosphate your body absorbs from food. Without enough of it, calcium passes through your digestive tract unused, no matter how much dairy or fortified food you eat. This is why prolonged deficiency leads to soft, weak bones in adults and rickets in children. Your skin produces vitamin D when exposed to sunlight, but people who live in northern latitudes, have darker skin, or spend most of their time indoors often fall short. Fatty fish, egg yolks, and fortified milk are dietary sources, though supplementation is common because few foods contain meaningful amounts.
Vitamin B12: Nerve and Blood Cell Builder
Vitamin B12 keeps two critical systems running: your nervous system and your blood supply. It helps maintain the protective coating around nerve fibers, and it’s essential for producing healthy red blood cells and synthesizing DNA in every cell of your body.
When B12 levels drop, the consequences show up in both systems. Red blood cells become abnormally large and inefficient, a condition called megaloblastic anemia that causes fatigue, weakness, and shortness of breath. Nerve damage can follow, producing numbness or tingling in the hands and feet, difficulty walking, memory problems, and mood changes. If the deficiency persists long enough, some of that nerve damage becomes permanent.
B12 is found almost exclusively in animal products: meat, fish, eggs, and dairy. This makes vegans and strict vegetarians especially vulnerable to deficiency, along with older adults whose stomachs produce less of the acid needed to release B12 from food. People who’ve had weight-loss surgery or who take certain acid-reducing medications are also at higher risk. For these groups, a supplement or fortified foods like nutritional yeast and plant milks can fill the gap reliably.
Vitamin C: Tissue Repair and Immune Support
Vitamin C is the body’s primary tool for building and repairing connective tissue. It serves as a required cofactor for the enzymes that construct collagen, the protein that holds together your skin, blood vessels, tendons, and bones. Without it, collagen production stalls. That’s exactly what happens in scurvy: blood vessels weaken, wounds stop healing, gums bleed, and joints swell. Scurvy is rare today, but subclinical deficiency is not. A 2024 modeling study published in The Lancet Global Health estimated that 53% of the world’s population doesn’t consume enough vitamin C.
Beyond tissue repair, vitamin C is a potent antioxidant that directly supports immune cells. It stimulates the production and function of white blood cells, particularly neutrophils (the first responders to infection) and lymphocytes (the cells that coordinate targeted immune attacks). White blood cells actually stockpile vitamin C at concentrations far higher than what’s found in the bloodstream, using it to protect themselves from the oxidative damage they generate while killing pathogens. Vitamin C also enhances the ability of neutrophils to migrate toward infection sites and destroy microbes once they arrive.
Fruits and vegetables are the best sources: citrus, bell peppers, strawberries, broccoli, and kiwi are all rich in it. One practical detail worth knowing is that your body absorbs 80 to 100% of vitamin C at doses up to about 200 mg, but absorption drops below 50% at doses above 1,000 mg. So spreading your intake across meals is more efficient than taking one large supplement. Studies have also found no difference in absorption between synthetic vitamin C supplements and the vitamin C found naturally in food, so either source works equally well.
How to Tell if You’re Low
Deficiency symptoms overlap enough to make self-diagnosis unreliable. Fatigue, for instance, can signal low levels of any of these three vitamins. A few patterns are more distinctive: tingling or numbness in the extremities points toward B12, frequent infections and slow wound healing suggest vitamin C or D, and bone pain with muscle weakness is a hallmark of vitamin D deficiency.
A standard blood test can measure levels of all three. If you eat a varied diet that includes animal products, fruits, vegetables, and some sun exposure, you’re likely covering your bases. The groups most at risk for gaps are vegans (B12), people with limited sun exposure (D), and those who eat very few fruits and vegetables (C). If any of those descriptions fit you, targeted supplementation is a straightforward fix, and the absorption data shows that common over-the-counter supplements work just as well as food sources for these particular vitamins.
