What Causes Mast Cell Tumors in Dogs: Breed, Age & More

Mast cell tumors in dogs arise from a combination of genetic mutations, breed-related predisposition, and aging, though no single definitive cause has been identified. They are one of the most common skin tumors in dogs, and understanding the known risk factors can help you recognize which dogs are most vulnerable and why these tumors develop.

How Normal Immune Cells Become Tumors

Mast cells are a normal part of your dog’s immune system. They live in skin, the lining of the gut, and the airways, where they respond to allergens, parasites, and injuries by releasing chemicals that trigger inflammation and healing. A mast cell tumor forms when these cells accumulate mutations that cause them to multiply uncontrollably instead of performing their usual immune functions.

The most well-understood mutation involves a protein on the surface of mast cells called KIT, which acts like a growth switch. In healthy cells, this switch turns on only when the body signals that more mast cells are needed. In roughly 20 to 30 percent of canine skin mast cell tumors, a mutation in the gene for this protein leaves the switch permanently stuck in the “on” position, driving constant cell division. A second, less common mutation in the same gene appears in about 2 to 5 percent of cases but doesn’t seem to affect how aggressive the tumor becomes.

Beyond that single gene, researchers have found at least 19 other genes with altered activity levels in aggressive, metastasizing tumors compared to benign ones. Specific small molecules that regulate gene expression have also been linked to whether a tumor spreads. In short, mast cell tumors aren’t caused by one broken gene. They result from a cascade of molecular changes that accumulate over time.

Breed Is the Strongest Known Risk Factor

Certain breeds develop mast cell tumors at dramatically higher rates than the general dog population. A large epidemiological study of insured dogs in Sweden found that Dogo Argentinos had 30 times the risk of the average dog, and Boxers had nearly 10 times the risk. Shar-Peis carry an especially concerning predisposition: while their overall tumor risk is elevated, their risk of dying from a mast cell tumor was over 50 times higher than average.

Other breeds consistently overrepresented in mast cell tumor diagnoses include Pugs, French Bulldogs, Golden Retrievers, Labrador Retrievers, Boston Terriers, and Bulldogs. The fact that these breeds cluster into a few genetic lineages strongly suggests inherited traits play a central role, even though specific genetic markers beyond the KIT mutation haven’t been pinpointed for most breeds.

Notably, some of the high-risk breeds tend to be diagnosed at younger ages. Pugs, Boxers, French Bulldogs, and Shar-Peis all show earlier onset compared to other breeds, which reinforces the idea that their genetic background accelerates the process.

Age and the Role of Time

Mast cell tumors are primarily a disease of adult and older dogs. The risk increases with each year of life, and older dogs are also more likely to develop higher-grade, more aggressive tumors. This pattern fits what we know about cancer biology in general: the longer cells divide over a lifetime, the more opportunities mutations have to accumulate. DNA repair mechanisms also become less efficient with age, allowing damaged cells to persist rather than being eliminated.

For high-risk breeds diagnosed young, inherited mutations likely give the process a head start, meaning fewer additional mutations are needed before a tumor forms.

Chronic Inflammation as a Possible Trigger

The exact cause of mast cell tumors remains unknown in the majority of cases, but chronic inflammation is one of the leading hypotheses. Since mast cells are already central players in the inflammatory response, prolonged activation from skin irritation, allergies, or other ongoing immune stimulation could theoretically push these cells toward uncontrolled growth. Alterations in tumor suppressor pathways (the body’s built-in brakes against cancer) and disrupted cell-cycle regulation have also been implicated.

This connection hasn’t been proven as a direct cause-and-effect relationship in dogs, but the biological plausibility is strong. Mast cells that are chronically activated divide more often, and more division means more chances for a copying error in their DNA.

Sex Does Not Appear to Be a Major Factor

Whether male or female dogs are more prone to mast cell tumors is still debated. Overall incidence is nearly identical between the sexes, at roughly 3 cases per 10,000 dogs annually for both males and females. One subtype, subcutaneous mast cell tumors (those forming under the skin rather than within it), may show a slight female predisposition, but the data is inconsistent across studies. For practical purposes, sex alone is not a useful predictor of risk.

Where the Tumor Appears Matters

While tumor location isn’t a cause, it does influence how the disease behaves once it develops. Mast cell tumors on the nail bed, muzzle, or inside the mouth tend to be more aggressive than those on the trunk or limbs. Dogs with mast cell tumors in internal organs face a significantly worse prognosis than those with skin tumors. Previously, tumors in the groin and perineal area were considered higher risk, but more recent studies suggest that may not hold true.

How Tumors Are Graded

When a mast cell tumor is removed and examined under a microscope, a pathologist assigns it a grade that reflects how likely it is to spread. The most widely used system classifies tumors as either low-grade or high-grade based on how abnormal the cells look: how often they’re actively dividing, whether they have multiple or oddly shaped nuclei, and whether their nuclei are unusually large. Low-grade tumors are far more common and often curable with surgery alone. High-grade tumors carry a much higher risk of spreading to lymph nodes and organs.

Testing the tumor for the KIT gene mutation can add another layer of information. Dogs whose tumors carry the mutation in the most common location face higher rates of metastasis and death, but those same tumors also tend to respond well to targeted drug therapies that block the overactive growth signal. This makes genetic testing both a prognostic tool and a guide for treatment decisions.

Putting the Pieces Together

No single environmental toxin, diet, or lifestyle factor has been proven to cause mast cell tumors in dogs. The strongest risk factors are ones you can’t control: your dog’s breed, their genetic makeup, and their age. Chronic inflammation may contribute, but the evidence is circumstantial rather than definitive. For owners of high-risk breeds, the most practical takeaway is vigilance. Any new lump or bump, especially one that changes size, becomes red, or seems itchy, warrants a veterinary exam. Mast cell tumors are among the most treatable cancers in dogs when caught early, particularly when they’re low-grade and haven’t spread beyond the skin.
