5-HTP (5-hydroxytryptophan) is a naturally occurring amino acid that your body uses as a building block for serotonin, the brain chemical that regulates mood, sleep, and appetite. It’s sold as a dietary supplement, typically extracted from the seeds of a West African plant called Griffonia simplicifolia, and is most commonly taken for depression, insomnia, and weight management.
How 5-HTP Works in Your Body
Your body normally makes 5-HTP on its own as part of a two-step process. First, it converts tryptophan (an amino acid you get from food) into 5-HTP. Then it converts 5-HTP into serotonin. That first step, tryptophan to 5-HTP, is the bottleneck. It’s slow, and it limits how much serotonin your body can produce at any given time.
When you take 5-HTP as a supplement, you skip that bottleneck entirely. The 5-HTP crosses from your bloodstream into your brain, where an enzyme quickly converts it into serotonin. This is also what distinguishes 5-HTP from L-tryptophan supplements: tryptophan still has to go through that slow conversion step, and much of it gets diverted to other uses in the body before it ever becomes serotonin. 5-HTP is one step closer to the finish line.
Because serotonin is also a precursor to melatonin (the hormone that regulates your sleep-wake cycle), raising serotonin levels with 5-HTP can indirectly support melatonin production as well.
5-HTP for Depression and Mood
The most common reason people take 5-HTP is to improve mood. A Cochrane review, the gold standard for evaluating medical evidence, found that 5-HTP and tryptophan were better than placebo at alleviating depression, with the magnitude of benefit appearing roughly similar to that of SSRIs and tricyclic antidepressants. However, the reviewers were careful to note that the evidence was not high enough quality to be conclusive. The clinical trials were small, and the body of research behind prescription antidepressants is far more rigorous.
That said, the Cochrane authors specifically noted that in cases of mild depression, where a person prefers not to take a prescription antidepressant, 5-HTP may be worth considering as an alternative. The side effects reported in trials were relatively mild: dizziness, nausea, and diarrhea were the most common.
5-HTP for Sleep
A 2024 randomized controlled trial tested 5-HTP supplementation in older adults over 12 weeks. The supplement raised blood serotonin levels and showed favorable effects on certain sleep quality measures. The most notable finding was that 5-HTP helped poor sleepers more than people who already slept reasonably well. Poor sleepers who took 5-HTP shaved nearly three minutes off the time it took them to fall asleep, a statistically significant improvement, while good sleepers saw little change.
Three minutes may sound modest, but the broader pattern mattered: the benefit showed up specifically in the people who needed it most. The connection between 5-HTP, serotonin, and melatonin production provides a plausible biological explanation for why it helps with sleep, though larger studies are still needed.
5-HTP for Appetite and Weight
Serotonin plays a role in satiety, which is why 5-HTP is sometimes marketed for weight loss. The theory is straightforward: more serotonin means you feel fuller sooner and eat less. Some older studies supported this idea, but more recent research has been less convincing. An 8-week randomized controlled trial found no changes in total calorie intake or macronutrient consumption between the group taking 5-HTP and the group taking a placebo. Participants were tracking their food intake throughout the study, and neither group ate meaningfully differently.
The appetite-suppression claim isn’t entirely baseless, but the current evidence doesn’t strongly support buying 5-HTP specifically for weight loss.
Side Effects and Safety Concerns
At typical supplement doses, 5-HTP is generally well tolerated. The most common side effects are gastrointestinal: nausea, diarrhea, and stomach discomfort. These tend to be mild and often improve after the first few days.
The more serious concern is serotonin syndrome, a potentially dangerous condition caused by too much serotonin in the body. Symptoms include rapid heart rate, agitation, hyperactive reflexes, and in severe cases, high fever and seizures. This risk is highest when 5-HTP is combined with medications that also raise serotonin levels. The two most important drug classes to be aware of are SSRIs (commonly prescribed antidepressants like fluoxetine and sertraline) and MAOIs (an older class of antidepressants). Animal studies have shown that combining 5-HTP with either of these drug classes can produce a dangerous serotonin reaction, and case reports in humans confirm the risk. If you take any medication that affects serotonin, 5-HTP is not safe to add on your own.
The Peak X Contamination Issue
In the late 1990s and early 2000s, researchers identified a contaminant called “Peak X” in some commercial 5-HTP products. Chemical analysis determined that one of these contaminants was a compound with potential neurotoxic properties. It was found in case-implicated 5-HTP samples as well as six over-the-counter products tested at the time, at levels ranging from 0.5% to over 10% relative to the contaminating compound. This raised concerns about manufacturing purity. While production standards have improved since then, 5-HTP is still sold as a dietary supplement, which means it is not subject to the same rigorous testing as prescription medications. Choosing products from manufacturers that perform third-party testing is one way to reduce this risk.
Common Doses
Most 5-HTP supplements are sold in 50 mg or 100 mg capsules. Clinical trials have used a wide range of doses, but most fall between 100 mg and 300 mg per day. Some people start at 50 mg to assess tolerance and increase gradually. Taking it with food can help reduce nausea. For sleep purposes, it’s typically taken in the evening; for mood support, it may be split across the day.
Because 5-HTP is classified as a supplement rather than a drug, there are no officially established dosing guidelines. The amounts used in research vary significantly depending on the condition being studied, and the optimal dose for any individual isn’t well defined by current evidence.
