A maternity test is a DNA test that confirms whether a woman is the biological mother of a child. It works the same way as a paternity test: a lab compares genetic markers in the mother’s DNA to those in the child’s DNA to determine if they share a biological parent-child relationship. While paternity tests get far more attention, maternity tests serve important roles in immigration, surrogacy, adoption, fertility treatments, and cases where hospital mix-ups are suspected.
How a Maternity Test Works
Every child inherits half their DNA from their biological mother and half from their biological father. A maternity test looks for shared genetic markers, which are specific characteristic patterns in DNA sequences, between the woman being tested and the child. The industry standard is to analyze at least 16 genetic markers, though some tests examine more for greater precision.
Results are reported as a probability of maternity. A result of 99.9% means the woman is almost certainly the biological mother. A result of 0% means there is no genetic match and she is not the biological parent. There is no middle ground in practice: the result either confirms or excludes the relationship.
Why Someone Would Need One
Maternity testing sounds unusual at first. After all, the woman who gives birth typically knows she’s the mother. But several real situations make a maternity test necessary or valuable.
Immigration cases: When a mother is sponsoring a child for an immigrant visa and no other credible documentation exists to prove the relationship, the U.S. Department of State may request DNA testing. Maternity is one of the commonly tested relationships alongside paternity and sibling connections.
IVF and fertility treatment verification: Embryo mix-ups in fertility clinics, while rare, do happen. When an IVF clinic discovers any discrepancy in its records, guidelines recommend offering genetic testing to confirm correct parentage. Some clinics have tested all embryos with even minor record inconsistencies at their own expense to ensure the right embryos went to the right families.
Surrogacy and adoption: In gestational surrogacy, where the surrogate carries an embryo created from another woman’s egg, a maternity test can confirm the intended mother’s genetic connection to the baby. In adoption cases, a birth mother or an adoptee may later seek DNA confirmation of their biological relationship.
Suspected hospital newborn mix-ups: Though modern hospital protocols have made this extremely rare, families who suspect a mix-up at birth can use maternity testing to get a definitive answer.
The Testing Process
The physical process is simple and painless. Both the woman and child provide DNA samples using a cheek swab. A foam-tipped applicator is rubbed along the inside of each cheek for about 10 seconds, collecting cells from the cheek lining. The swab is then either pressed onto a special collection card or placed back into sterile packaging and sent to a lab. No blood draw is needed, and the whole collection takes just a few minutes.
For young children and infants, the process is the same. The swab is gentle enough for a newborn.
Legal Tests vs. At-Home Tests
There are two categories of maternity tests, and the distinction matters depending on what you need the results for.
- At-home (informational) tests let you collect cheek swab samples in your own home using a kit with step-by-step instructions. You mail the samples to the lab in a prepaid envelope. These results satisfy personal curiosity but carry no legal weight because there’s no way to verify who actually provided the samples.
- Legal tests require sample collection at a certified facility where staff verify each person’s identity and maintain a documented chain of custody. This means every step, from who provided the sample to how it was handled and transported, is recorded and verifiable. Courts, immigration agencies, and government bodies only accept results from legal tests.
If you need the results for an immigration application, custody case, or any other legal proceeding, you must use a legal test from the start. An at-home test cannot be upgraded to legal status after the fact.
Accuracy and Reliability
Maternity DNA tests are extremely accurate. When a biological relationship exists, accredited labs report a probability of 99.9% or higher. When no relationship exists, the result is 0%. The science behind the comparison is the same whether you’re testing for maternity or paternity.
Accuracy improves when more genetic markers are analyzed. The baseline standard of 16 markers is sufficient for a reliable result, but some labs test additional markers to increase confidence, particularly in complex cases where close biological relatives (like sisters) might both be potential mothers. If you’re choosing an at-home kit, look for one that tests at least the standard 16 markers.
Cost and Turnaround Time
At-home maternity tests generally cost between $100 and $300, depending on the lab and the number of markers analyzed. Legal tests run higher, typically $300 to $500 or more, because of the added requirements for identity verification and chain-of-custody documentation. In immigration cases, the testing lab may need to coordinate sample collection across multiple countries, which can add both time and cost.
Most labs return results within 3 to 5 business days after receiving samples, though legal tests and international cases can take longer due to shipping and administrative steps.
