What Is a NIPP Test and How Does It Work?

A NIPP test, or noninvasive prenatal paternity test, is a DNA-based paternity test that can determine the biological father of a baby during pregnancy using only a blood draw from the mother and a cheek swab from the potential father. It is 99.9% accurate and can be performed as early as eight weeks into pregnancy. Unlike older methods that required a needle into the uterus, a NIPP test carries no risk of miscarriage because it never touches the pregnancy itself.

How the Test Works

Starting around the sixth week of pregnancy, small fragments of the baby’s DNA begin circulating in the mother’s bloodstream. This happens naturally as the placenta develops: cells break down and release DNA into the mother’s blood. The amount of fetal DNA in the mother’s blood increases as the pregnancy progresses, eventually making up anywhere from 2% to 20% of all the free-floating DNA in her plasma.

A NIPP test captures that fetal DNA from a standard blood sample drawn from the mother’s arm. The lab then compares the baby’s genetic markers to a cheek swab taken from the potential father. If the DNA profiles match, paternity is confirmed. If they don’t, the tested man is excluded as the father. No sample is needed from the baby directly, and nothing invasive is done to the pregnancy.

When You Can Take It

Most labs require you to be at least eight weeks pregnant before performing a NIPP test. At that point, enough fetal DNA is present in the mother’s blood for the lab to isolate and analyze it reliably. Testing earlier than this increases the chance of an inconclusive result simply because there isn’t enough fetal DNA to work with yet.

The test can be done at any point after that eight-week mark, all the way through delivery. Fetal DNA levels rise steadily throughout pregnancy, so later testing generally produces stronger results. That said, eight weeks is considered reliable enough for most people who need answers early.

Why It Replaced Invasive Methods

Before NIPP testing existed, the only way to determine paternity during pregnancy was through invasive procedures like amniocentesis or chorionic villus sampling (CVS). Both involve inserting a needle or catheter into the uterus to collect fetal cells directly. These procedures carry a small but real risk of miscarriage, infection, and other complications.

A NIPP test eliminates those risks entirely. The mother gives a blood sample from her arm, and the potential father provides a cheek swab. There is no contact with the uterus, no sedation, and no recovery time. For most people, the entire sample collection takes less than 15 minutes.

Accuracy and Reliability

NIPP tests are 99.9% accurate when performed correctly with sufficient fetal DNA in the sample. Results typically report a “probability of paternity” that is either extremely high (confirming paternity) or zero (excluding the tested man entirely).

The main reason a test can fail or come back inconclusive is low fetal fraction, meaning the baby’s DNA makes up too small a percentage of the total free-floating DNA in the mother’s blood. Most labs require at least 4% fetal DNA in the sample to report a confident result. Samples below that threshold are considered unreliable because the lab can’t distinguish the baby’s genetic markers clearly enough.

Two factors most commonly cause low fetal fraction. The first is higher maternal BMI. Research shows that BMI is the single strongest predictor of a failed test: in one study, 74% of samples with inadequate fetal DNA came from clinically obese patients, compared to 27% of samples that had adequate levels. The second factor is early gestational age. Testing very close to the eight-week cutoff leaves less margin for fetal DNA accumulation. If a test fails for either reason, retesting a few weeks later often solves the problem as fetal DNA levels continue to rise.

What Samples Are Needed

The test requires two samples:

  • From the mother: A blood draw from the arm, collected into specialized tubes that preserve the free-floating DNA in her plasma.
  • From the potential father: A simple cheek swab, where a soft brush is rubbed along the inside of the cheek to collect cells containing his DNA.

Both samples are sent to the lab together. Results typically come back within one to two weeks, depending on the lab.

Legal Admissibility

Not all NIPP tests hold up in court. The difference comes down to chain of custody. A legally admissible test requires that sample collection be witnessed and documented by a neutral third party, usually at a clinic or lab facility. Every step, from identification of the people being tested to sealing and shipping the samples, is tracked to prove no tampering occurred.

An at-home or “peace of mind” version of the test uses the same science and delivers the same level of accuracy, but because no one independently verifies who provided the samples, the results generally cannot be used in custody cases, child support hearings, or other legal proceedings. If you need results that will stand up in court, make sure you specifically request a legal, chain-of-custody test and have samples collected at an approved facility.

Common Reasons People Get Tested

NIPP testing is used in a range of situations. Some people simply want certainty about paternity before the baby is born so they can plan accordingly. Others need early answers for legal or forensic reasons. In cases involving sexual assault, the test allows judicial authorities and victims to establish paternity within weeks of pregnancy rather than waiting until after delivery, which can be critical for legal proceedings.

Because the test poses no risk to the pregnancy and requires only routine sample collection, the barrier to testing is low. The primary consideration for most people is cost, which typically ranges from several hundred to over a thousand dollars depending on the lab, turnaround time, and whether a legal chain of custody is included. Insurance rarely covers prenatal paternity testing.
