What Is Dissolution Rate and Why Does It Matter?

Dissolution rate is how fast a solid disappears into a liquid, like sugar into tea or a tablet into stomach fluid. It is a separate question from how much of the solid can dissolve at all.1ClinicalGate. Dissolution and solubility That difference matters because speed often decides the outcome: whether a medicine works on time, whether a soft drink wears at your teeth, or whether a layer of rock turns into a cave.

Speed Versus Amount, What Dissolution Rate Measures

Solubility is the most of a substance a liquid can hold before it refuses to take any more. Dissolution rate is how quickly the solid gets there, measured as the amount dissolved over time. The two aren’t necessarily linked, so a substance can dissolve slowly and still dissolve a lot in the end.2ClinicalGate. Dissolution and solubility A sugar cube and the same weight of loose sugar can hold the same total amount in a cup, yet the loose sugar vanishes in seconds while the cube lingers. Either way, once the water is full, it stops dissolving no matter how long you wait.

A label saying a substance is soluble describes the end state, not the journey, so it can’t tell you whether the wait is seconds or days. Two powders with the same ability to dissolve can differ enormously in waiting time because of how they are packed, shaped, or sized.

Dissolving happens at the solid’s surface, where molecules leave into a thin layer of liquid clinging to it. The rate depends on how much surface remains and on the gap between the concentration in that layer and in the surrounding liquid.3PubMed Central. Dissolution Kinetics of a BCS Class II Active Pharmaceutical Ingredient: Diffusion-Based Model Validation and Prediction The layer touching the solid quickly becomes crowded with dissolved material, and fresh liquid has to replace it for dissolving to continue.

As the liquid fills up, the gap shrinks and dissolving slows. So dissolving usually starts fast and tapers off near the limit. If you could measure the sugar left in a stirred cup every few seconds, most of the loss would happen early, and the last few grains would take disproportionately long to vanish. Lab tests of tablets show the same shape: a chart of percent dissolved against time climbs steeply, then flattens.

What Makes Things Dissolve Faster or Slower

Dissolving speed rises with the exposed surface, the total outer area touching the liquid.4Ardena. The Science of Solubility: Particle Size Reduction Techniques Grinding a solid into smaller pieces exposes more surface and speeds dissolving.5Ardena. The Science of Solubility: Particle Size Reduction Techniques Instant coffee granules disappear faster than a solid lump would.

The effect can be dramatic. In one test, coenzyme Q10 crystals about 80 nanometres across dissolved 65% in a day, versus 13% for 120-nanometre crystals.6PubMed Central. Effect of particle size on solubility, dissolution rate, and oral bioavailability: evaluation using coenzyme Q10 as naked nanocrystals Larger crystals and ordinary powder barely dissolved at all in the same liquid.7PubMed Central. Effect of particle size on solubility, dissolution rate, and oral bioavailability: evaluation using coenzyme Q10 as naked nanocrystals But smaller isn’t always better. Some materials, such as silica particles, don’t speed up as much as surface predicts when they shrink.8PubMed. The dissolution rates of SiO2 nanoparticles as a function of particle size Very small particles can also clump together and lose their advantage, so size helps only until other limits take over.

Stirring thins the crowded layer of liquid around the solid, so dissolving speeds up.9PubMed. Effect of hydrodynamic environment on tablets dissolution rate In tablet tests, spinning four times faster made the surrounding layer about six times thinner and dissolving roughly five to six times faster.10PubMed. Effect of hydrodynamic environment on tablets dissolution rate How the liquid is stirred, not just how fast, changes the result too.11PubMed. Effect of hydrodynamic environment on tablets dissolution rate

Stirring helps most when that layer is the bottleneck. Once the layer is already very thin, stirring harder gains little, and speed is limited by how fast molecules leave the solid itself. A gentle swirl that reaches every grain can beat a hard stir that leaves some grains sitting in still liquid.

Temperature is another lever. Warmer liquid generally speeds dissolving because molecules move faster and carry dissolved material away from the surface more quickly. Heat often raises how much can dissolve as well, so it changes both the speed and the limit, which is why sugar goes into hot tea far more readily than iced tea. It isn’t universal: a few substances dissolve less in warmer water, so “hotter is faster” is a common pattern, not a law.

The solid’s own structure matters too. The same chemical can pack into different crystal shapes, and these dissolve differently.12PubMed Central. Impact of crystal polymorphism of rifaximin on dissolution behavior Four crystal forms of the antibiotic rifaximin ranged from about 3.5 to 8.4 micrograms per millilitre in how much dissolved.13PubMed Central. Why Dissolution Rate Matters for Your Medicine

A swallowed tablet must release its drug, and the drug must dissolve, before the gut can absorb it. That makes dissolving often the slowest step in getting a drug from a pill into the body.17PubMed. Drug dissolution: significance of physicochemical properties and physiological conditions Drugs that dissolve poorly in water, below about 100 micrograms per millilitre, often run into this limit.18PubMed. Drug dissolution: significance of physicochemical properties and physiological conditions Speeding dissolving, for example by shrinking particles, can raise how much drug the body takes in.19Ardena. The Science of Solubility: Particle Size Reduction Techniques

The time pressure is real. A pill spends only a few hours moving through the digestive tract, and absorption mostly happens in the intestine. The drug has to be in solution while it passes the stretch of gut where it can be absorbed. Whatever hasn’t dissolved by the time it moves past may leave the body in stool unused. That is why poorly dissolving drugs are hard to make: makers grind them, change their crystal form, or add helper ingredients to fix the problem.

Speed stops being the bottleneck at some point. Once a drug dissolves quickly enough, or a larger dose is given, the limit shifts to how much can dissolve at all.20PubMed. Rate-limiting steps of oral absorption for poorly water-soluble drugs in dogs; prediction from a miniscale dissolution test and a physiologically-based computer simulation Past that point, faster dissolving no longer helps absorption.

Regulators sort drugs into four groups by how well they dissolve and how easily they pass through the gut wall.21PubMed Central. Prediction of solubility and permeability class membership: provisional BCS classification of the world’s top oral drugs The sorting predicts which step, dissolving or passing through, will slow absorption.22PubMed Central. Prediction of solubility and permeability class membership: provisional BCS classification of the world’s top oral drugs A drug that does both well is rarely held back by its formulation, so small manufacturing changes matter less. Common drugs such as ibuprofen and diclofenac sit in the group that passes easily but dissolves poorly.23PubMed Central. Dissolution Kinetics of a BCS Class II Active Pharmaceutical Ingredient: Diffusion-Based Model Validation and Prediction For these, dissolving is the main hurdle, so how the pill is made has a large say in how fast it works.

For you, this shows up in a few ways. Two brands of the same drug must show similar dissolving behaviour, among other tests, to be considered interchangeable. Fast-acting forms like liquid gels are designed to dissolve quickly. Crushing or splitting a pill changes how it dissolves and can be unsafe for some, so ask a pharmacist before altering any tablet.

Slow-Release Pills Use Dissolving on Purpose

Regular tablets aim to dissolve fast and fully. Slow-release tablets aim for steady dissolving over many hours, which often allows fewer doses per day. Some do this by mixing the drug with a material that soaks up water and forms a gel layer on the surface.24PubMed Central. Effect of Drug Solubility on Polymer Hydration and Drug Dissolution from Polyethylene Oxide (PEO) Matrix Tablets Water must work its way into the tablet, and the drug must seep out through the gel, which delays release.25PubMed Central. Effect of Drug Solubility on Polymer Hydration and Drug Dissolution from Polyethylene Oxide (PEO) Matrix Tablets

How fast the drug leaves depends on how well it dissolves, how thick the gel gets, and how much gel-forming material is used.26PubMed Central. Effect of Drug Solubility on Polymer Hydration and Drug Dissolution from Polyethylene Oxide (PEO) Matrix Tablets Changing filler ingredients, such as lactose or starch, also speeds or slows release.27PubMed. Studies on drug release kinetics from ibuprofen-carbomer hydrophilic matrix tablets: influence of co-excipients on release rate of the drug

That design is why you shouldn’t crush these pills. Crushing destroys the gel barrier, so the whole dose can dissolve at once, which can mean too much drug too quickly. Labels often say “swallow whole” for this reason.

Makers check dissolving in standard lab vessels with a stirring paddle or spinning basket.28Dissolution Technologies. History and Evolution of the Dissolution Test A tablet goes into fluid kept at body temperature while the paddle turns at a set speed, and samples are taken at planned times to see what percent has dissolved. For slow-release tablets the result should be a gradual rise over hours, and drifting outside the expected range signals a problem. Regulators use these tests to judge whether a pill will behave in the body as expected.29PubMed. Modeling and comparison of release profiles: Effect of the dissolution method

Soft Drinks and Your Tooth Enamel

Enamel erosion is acid slowly dissolving the mineral surface of teeth. In lab tests, every soft drink tested caused some enamel loss.30PubMed. Impact of modified acidic soft drinks on enamel erosion Original Sprite and orange juice dissolved enamel more than other drinks, and original Coca-Cola least.31PubMed. Impact of modified acidic soft drinks on enamel erosion

Acidity alone doesn’t explain the differences. How much a drink harms enamel depends on what acids it contains and how much buffering it has. Some acids, such as citric acid, bind calcium and pull it from enamel, adding damage beyond simple acidity. That is why drinks with similar acidity can still differ in how much enamel they dissolve.

Saliva helps teeth recover. It gradually neutralizes acid and supplies minerals that can partly re-harden softened enamel. Recovery takes time, so frequent acid exposure leaves less opportunity to repair, and the number of exposures across the day matters, not only the total amount drunk. Sipping all day keeps teeth bathed in acid longer than drinking quickly. Rinsing with water afterward dilutes acid, and waiting before brushing avoids scrubbing softened enamel. A straw reduces contact with the front teeth.

Shells, Caves and Climate

Carbon dioxide from the air dissolves into seawater and reacts with water to form an acid. As more is released into the air, more enters the ocean, shifting the water toward acidity. That is a dissolving process too, a gas dissolving into a liquid, and the more acidic seawater speeds the dissolving of the chalky mineral in shells.32EDIS. Ocean Acidification: Calcifying Marine Organisms Animals such as snails, oysters, and sea butterflies build shells from this mineral.

In one study, snails’ shells lost weight under higher carbon dioxide because existing shell dissolved, not because they built less.33PubMed Central. Elevated CO2 affects shell dissolution rate but not calcification rate in a marine snail Thin shells of Antarctic sea creatures showed clear dissolving within two to five weeks in strongly acidic water.34Antarctic Science. Rapid dissolution of shells of weakly calcified Antarctic benthic macroorganisms indicates high vulnerability to ocean acidification Even if an animal keeps building, it may lose shell as fast as it adds it, leaving a thinner or weaker covering. Thin shells protect less against predators, which is how shell dissolving can reach up the food web.

Three Years of Field Trials Indicate a Sustained Enhanced Rock Weathering Signal with Limited CO(2) Removal Crushing exposes more surface, as with grinding a drug, so dissolving should be faster. But three years of vineyard trials found removal 10 to 30 times lower than some earlier estimates.40PubMed.
