The active ingredient in Vyvanse has a half-life of roughly 12 hours. That means it takes about 12 hours for your body to clear half of the drug’s active form from your bloodstream, and around two and a half days for it to wash out almost entirely. But the full picture is more nuanced than a single number, because Vyvanse doesn’t arrive in your system as a ready-to-go stimulant.
Why Vyvanse Has Two Half-Lives
Vyvanse is a prodrug, meaning the pill itself isn’t active. What you swallow is lisdexamfetamine, a molecule that your body has to convert before it does anything. That conversion happens inside red blood cells, where enzymes clip off an amino acid and release dextroamphetamine, the compound that actually treats ADHD and binge eating disorder.
Lisdexamfetamine itself is processed quickly. It reaches its peak concentration in your blood about one hour after you take it, then drops off fast as red blood cells steadily break it down. The dextroamphetamine it produces peaks later, around 3.5 to 4.4 hours after the dose depending on the formulation (capsule vs. chewable tablet). That staggered release is by design: it creates a smoother, longer-lasting effect than taking dextroamphetamine directly.
The 12-hour half-life applies to dextroamphetamine, the part that’s actually working in your brain. This is the number that matters for how long you feel the medication’s effects and how long it stays detectable.
How Long Vyvanse Stays in Your System
A drug is generally considered cleared after about five half-lives. With a 12-hour half-life, that puts full elimination at roughly 60 hours, or about two and a half days after your last dose. Here’s how the math works for a single dose:

Factors That Shorten or Extend It
That 12-hour figure is an average, and your actual clearance time can shift significantly based on a few variables.
Urine pH is the biggest one. Your kidneys excrete dextroamphetamine more efficiently when your urine is acidic and less efficiently when it’s alkaline. The range is dramatic: urinary recovery of amphetamine can be as low as 1% or as high as 75% depending on pH. In practical terms, this means a diet high in acidic foods or drinks (cranberry juice, high-protein meals, vitamin C supplements) can speed elimination, while alkaline conditions (from antacids, certain diets, or medications like sodium bicarbonate) can slow it down and effectively extend the drug’s duration.

How This Compares to Other ADHD Medications
Vyvanse’s 12-hour half-life is longer than immediate-release stimulants, which typically clear in 4 to 6 hours for dextroamphetamine formulations. The prodrug design is the reason: because red blood cells have to convert the molecule before it becomes active, the release is gradual and self-limiting. You can’t speed it up by crushing the pill or taking a larger dose, since the conversion process has a built-in ceiling based on how fast your red blood cells work.
This is also why Vyvanse tends to produce a smoother onset and offset than medications that deliver amphetamine directly. The tradeoff is that it takes longer to kick in (peaking around 3.5 to 4.4 hours versus about 1 to 2 hours for immediate-release options) and its effects can linger into the evening, which some people find helpful and others find disruptive to sleep.
