How Daxxify Outlasts Other Neurotoxins

All injectable neurotoxins work the same basic way: they block nerve signals to muscles, temporarily relaxing the contractions that cause wrinkles. What separates Daxxify is a proprietary peptide called RTP004 that acts like molecular glue. This 35-amino-acid peptide carries a strong positive charge that binds to the negatively charged surfaces of nerve cells, essentially anchoring the toxin at the injection site longer than competing products. The longer the toxin stays parked at the nerve terminal, the more of it gets absorbed into the nerve cell, extending the effect.

Every other neurotoxin on the market uses human serum albumin as a stabilizer. Daxxify replaces that with RTP004 and a surfactant, making it the first albumin-free formulation. This isn’t just a manufacturing distinction. The peptide’s stickiness to nerve terminals is the core reason Daxxify outlasts its competitors by months rather than days.

In the large Phase 3 clinical trial (SAKURA 3), about 34 to 38% of patients still showed meaningful improvement at week 24, a full six months after injection. Even at week 36 (nine months), roughly 8 to 12% of patients still had visible improvement. No other neurotoxin comes close to those numbers.

How the Standard Options Compare

Botox, Dysport, Xeomin, and Jeuveau all fall within a similar duration window of roughly 3 to 4 months. A comparative study evaluating all major neurotoxin types found that more than 50% of participants experienced wrinkle improvement for up to 20 weeks (about 5 months) with most formulations. The minimum effective duration across all types was at least 12 weeks.

Where they differ slightly is in how fast they kick in. Dysport tends to show visible results within 2 to 3 days, while Botox typically takes 7 to 10 days to reach full effect. But once both are working, their durations are comparable. Neither product offers a meaningful longevity advantage over the other.

The newest entry, Letybo (approved by the FDA in 2024), showed statistically significant results lasting up to 16 weeks in its primary clinical trial. That places it squarely in the same 3-to-4-month range as the others, not in Daxxify territory.

Why Xeomin May Last Longer Over Time

One factor that rarely comes up in duration comparisons is immune resistance. Your body can develop antibodies against neurotoxins after repeated injections, gradually weakening the effect. The culprit is the extra proteins that come packaged with the active toxin. Botox contains about 5 nanograms of clostridial protein per vial, and Dysport contains 12.5 nanograms.

Xeomin is manufactured with a double-filtration process that strips away all complexing proteins, leaving only the pure 150-kilodalton neurotoxin. Its protein load is just 0.6 nanograms per vial. Those complexing proteins, particularly a group called hemagglutinins, are known to stimulate immune cells. Removing them may reduce the risk of your body building resistance over years of treatment. What Actually Affects How Long Your Results Last

The product you choose matters, but it’s not the only variable. Several factors influence how quickly any neurotoxin wears off.

  • Metabolism: People with faster metabolisms, including those who exercise intensely and frequently, tend to break down neurotoxins more quickly. There’s no reliable way to change this, but it helps explain why your results might differ from a friend’s.
  • Treatment area: Muscles that you use constantly (like the ones between your eyebrows when you concentrate or squint) tend to metabolize the toxin faster than less active areas.
  • Dosing: Higher doses can modestly extend results, but clinical guidelines note that this effect is limited. Doubling the dose does not double the duration.
  • Zinc supplementation: A double-blind, placebo-controlled study found that taking 50 mg of zinc citrate with phytase (an enzyme that helps your body absorb zinc) before treatment increased the duration of effect by nearly 30% in 92% of subjects. Lower doses of zinc without phytase showed no benefit. Neurotoxins are zinc-dependent enzymes, so adequate zinc levels may help the toxin function more effectively once inside the nerve cell.

Daxxify costs more per unit, typically $15 to $25 compared to Botox’s $10 to $20. But because you need fewer treatments per year, the annual cost often works out to be roughly the same. With Botox, most people schedule 3 to 4 appointments per year. With Daxxify, that drops to 1 or 2.
